We have developed PotatoMASH (Potato Multi-Allele Scanning Haplotags), a novel low-cost, genome-scanning marker platform. designed panel of 339 multi-allelic regions placed at 1 Mb intervals throughout the euchromatic portion genome. These were assayed using multiplex amplicon sequencing approach, which allows for genotyping hundreds plants cost 5 EUR/sample. applied to population over 700 potato lines. obtained tetraploid dosage calls 2012 short haplotypes in 334 loci, ranged from 2 14 different per locus. The system was able diagnose presence targeted pest-resistance markers, detect quantitative trait loci (QTLs) by genome-wide association studies (GWAS) population, and track variation diploid segregating population. efficiently surveys genetic genome, can be implemented as single low-cost platform that will allow routine simultaneous application marker-assisted selection (MAS) other applications commercial breeding programmes.
